hCG contains an alpha subunit shared with LH, FSH and TSH, and a hormone-specific beta subunit. The two chains associate noncovalently; isolated subunits do not reproduce the receptor-binding activity of the assembled hormone. Multiple N- and O-linked glycans create charge and mass heterogeneity, so an approximate 36–40 kDa value is more informative than a single exact mass but still does not define a preparation. Urinary and recombinant hCG can have different glycoform profiles.

How Does HCG Work in Research Models?

hCG activates the luteinizing hormone/choriogonadotropin receptor (LHCGR), a Gs-coupled receptor that increases cAMP and drives steroidogenic signalling in appropriate gonadal cell models. Beta-arrestin, ERK and receptor-internalization responses can differ with ligand exposure and cellular context. Useful proximal measurements include cAMP, receptor occupancy and phosphorylation; steroid output is a later, cell-dependent response.

How Should a HCG Study Be Designed?

Report activity in calibrated units as well as mass and state the source—recombinant, urinary or other. Include an LHCGR-negative control, receptor antagonism or knockout, and LH as a comparator when ligand bias is relevant. Do not compare lots by nominal micrograms without accounting for specific activity.

How Should HCG Be Handled and Verified?

Preserve quaternary structure and glycans by avoiding vigorous mixing, repeated freeze-thaw and unqualified reducing conditions. Use low-binding consumables at dilute concentrations. How Can HCG Results Be Interpreted Without Overclaiming?

Begin interpretation with a falsifiable question and a predeclared primary readout. For HCG, chemical identity, target engagement and downstream phenotype are separate layers. A change in migration, viability or gene expression does not by itself prove the proposed molecular target. Report negative and null findings, not only the largest response. Show individual observations, biological replicates and uncertainty intervals, and identify whether replication means a new well, a new culture, a new animal or a new day.

A citable HCG result states the species, cell line or biochemical system; exact molecular form; concentration and exposure; comparator; assay method; and statistical unit. Confirm that vehicle, pH, osmolality or precipitation did not create the phenotype. Conclusions should stay at the level tested: in vitro activity is not evidence of organism-level efficacy, and an animal phenotype is not a human claim.
